The liver can experience damage induced by drug (drug induced liver injury). Soursop plants are widely used as medicines such as antioxidants, antitumors, antimicrobials and antiparasites. The aim of this study is to determine morphological changes of rat liver tissue given long-term soursop leaves extract preparation (Annona muricata). This research was an experimental in vivo study with a Randomized Complete Design against Wistar strain rats (Rattus norvegicus) which divided into four groups: positive control group (normal) and three treatment groups with dosage of 20 mg/kg body weight soursop leaf extract; 40 mg/kg body weight and 80 mg/kg body weight given over 60 days. All treatments are administered orally. After 60 days of treatment, the rats were sacrificed and the liver was taken, then the histopathologic preparations were made and viewed using a light microscope. The results of the study showed an increase in the number of swollen hepatocytes in the three treatment groups with the most significant concentration of 80 mg / kgBW, and the decrease of intact central vein without necrosis area in the three treatment groups with the most significant concentration was 40 mg / kgBW. Both illustrate the effects of injury on liver cells. The conclusions of this study indicate that the provision of long-term soursop leaves extract affects the increase in the number of hepatocytes swell and decrease the number of central vein intact. |

Hati dapat mengalami kerusakan yang dapat disebabkan oleh obat-obatan (drug induced liver injury). Tanaman sirak banyak di gunakan sebagia obat-obatan seperti antioksidan, antitumor, antimikroba dan antiparasit. Penelitian ini bertujuan untuk mengetahui perubahan morfologi jaringan hati tikus pada pemberian sediaan ekstrak daun sirsak (Annona muricata) jangka panjang. Penelitian ini merupakan penelitian eksperimental in vivo dengan desain penelitian Rancangan Acak Lengkap terhadap tikus jantan galur wistar (Rattus norvegicus) yang terbagi dalam empat kelompok, yaitu kelompok kontrol positif (normal) dan tiga kelompok perlakuan dengan dosis ekstrak daun sirsak 20 mg/kgBB; 40 mg/kgBB dan 80 mg/kgBB yang diberikan selama 60 hari. Semua perlakuan diberikan secara oral. Setelah diberi perlakuan 60 hari, tikus dikorbankan dan diambil organ hatinya, kemudin dibuat sediaan histopatologi dan dilihat menggunakan mikroskop cahaya. Hasil penelitian menunjukkan adanya peningkatan jumlah hepatosit yang membengkak pada ketiga kelompok perlakuan dengan konsentrasi paling signifikan adalah 80 mg/kgBB, dan penurunan jumlah central vein yang utuh tanpa area nekrosis pada ketiga kelompok perlakuan dengan konsentrasi paling signifikan adalah 40 mg/kgBB. Keduanya menggambarkan efek cedera pada sel hati. Kesimpulan penelitian ini menunjukkan bahwa pemberian ekstrak daun sirsak jangka panjang berpengaruh terhadap paningkatan jumlah hepatosit yang membengkak dan panurunan jumlah central vein yang utuh. |
