ABSTRACT: Staphylococcus epidermidis is one of the bacteria that cause infection. Star fruit leaves (Averrhoa bilimbi L.) is known to have antibacterial activity due to the presence of secondary metabolites. However, it is uncertain whether the compound that plays an active role in Averrhoa bilimbi L. as an antibacterial especially against Staphylococcus epidermidis. Therefore, study aimed to determine the compounds that act as antibacterial. Crude material was firstly stratified by maceration and using n-hexane, ethyl acetate and methanol as solvents. The extracts collected were then tested for antibacterial activity to Staphylococcus epidermidis at a concentration of 10.000; 20.000; 30.000; 40.000; and 50.000 ppm using the diffusion method for the well technique. The test results showed that the extracts of n-hexane, ethyl acetate, and methanol produced inhibitory diameters of 15.9 mm ± 0.9; 12.45 mm ± 2.72; and 14.2 mm ± 4.16 consecutively. n-hexane extract which showed the best activity was tested then by contact bioautography TLC to detect the presence of compounds that have antibacterial activity. However, clear areas and color patches have not been identified resulting from the appearance of alkaloid, flavonoid, and phenol patches. Based on the results of phytochemical screening, n-hexane extract contains alkaloid, polyphenolate, flavonoid, anthraquinone, tanin, monoterpene and sesquiterpene, triterpenoid and steroid, which have potency in antibacterial activity resulted.Keywords: Averrhoa bilimbi L, antibacterial, Staphylococcus epidermidis, Bioautography TLC. ABSTRAK: Staphylococcus epidermidis adalah salah satu bakteri penyebab infeksi. Daun belimbing manis (Averrhoa bilimbi L.) diketahui memiliki aktivitas sebagai antibakteri berdasarkan kandungan metabolit sekundernya. Namun belum dapat dipastikan senyawa apakah yang berperan aktif dalam Averrhoa bilimbi L. sebagai antibakteri khususnya terhadap Staphylococcus epidermidis. Oleh karena itu, penelitian ini bertujuan untuk menentukan senyawa yang berperan sebagai antibakteri. Simplisia dan ekstrak dengan metode ekstraksi bertingkat (maserasi) dan menggunakan pelarut n-heksana, etil asetat dan metanol. Ekstrak yang diperoleh selanjutnya diuji aktivitas antibakteri terhadap Staphylococcus epidermidis pada konsentrasi 10.000; 20.000; 30.000; 40.000; dan 50.000 ppm menggunakan metode difusi agar teknik sumuran. Hasil uji menunjukkan bahwa ekstrak n-heksana, etil asetat, dan metanol menghasilkan diameter hambat secara berurutan masing-masing sebesar 15,9 mm ± 0,9; 12,45 mm ± 2,72; dan 14,2 mm ± 4,16. Ekstrak n-heksana yang menunjukkan aktivitas paling baik selanjutnya dilakukan KLT bioautografi kontak untuk mendeteksi keberadaan senyawa yang memiliki aktivitas sebagai antibakteri. Namun, belum dapat teridentifikasi area jernih dan bercak warna yang dihasilkan dari penampak bercak alkaloid, flavonoid, dan fenol. Adapun berdasarkan hasil penapisan fitokimia, golongan senyawa yang terdeteksi pada ekstrak n-heksana adalah alkaloid, polifenolat, flavonoid, antrakuinon, tanin, monoterpen dan sesquiterpen, triterpenoid dan steroid, yang berpotensi dalam aktivitas antibakteri yang dihasilkan.Kata Kunci: Averrhoa bilimbi L, antibakteri, Staphylococcus epidermidis, KLT bioautografi. |
