Abstact. Antioxidant peptides are peptides with antioxidant activity that protects against free radicals by donating electrons to free radicals. One of the natural antioxidant peptides that researchers found was tetrapeptide (Ser-Pro-Lys-Thr) which was isolated from the fungus (Tolypocladium inflatum) located in the Norwegian soil and was found to have antioxidants activity. This research has succeeded in synthesizing tetrapeptide (Ser-Pro-Lys-Thr) using the solid phase peptide (SPPS) method using the Fmoc-amino acid strategy using 2-chlorotrityl chloride resin as a buffer and coupled with a coupling reagent, HOBt and DIPEA. The sample weight resulted was 111.4 mg which was then characterized using a mass spectrophotometer and it was obtained the peak of the fragment at m / z 431. RP-HPLC was used to obtain the purity of the tetrapeptide mixture which resulted peak retention time at 9,243 minutes. Antioxidant activity was tested using the DPPH method, the results obtained from tetrapeptide compounds was 45% inhibition concentration indicating the antioxidant activity of tetrapeptide compounds is weak.Keywords: Antioxidant, tertapeptide, solid phase peptide synthesis (SPPS)Abstrak. Peptida antioksidan merupakan peptida yang memiliki aktivitas antioksidan yang berperan untuk menangkal radikal bebas dengan cara mendonorkan elektronnya kepada radikal bebas. Salah satu peptida antioksidan alami yang telah ditemukan peneliti sebelumnya adalah tetrapeptida (Ser-Pro-Lys-Thr) yang diisolasi dari jamur (Tolypocladium inflatum) yang berada ditanah Norwegia dan dilaporkan memiliki aktivitas antioksidan. Pada penelitian ini telah berhasil disintesis tetrapeptida (Ser-Pro-Lys-Thr) dengan menggunakan metode sintesis peptida fase padat (SPPS) menggunakan stategi Fmoc-asam amino, penyangga yaitu resin 2-klorotritil klorida dan dikopling dengan reagen pengkopling diantaranya HBTU, HOBt dan DIPEA. Hasilnya didapat bobot sampel sebesar 111,4 mg yang kemudian dilakukan proses karakterisasi dengan menggunakan spektrofotometer massa didapat puncak fragmennya pada m/z 431 dan untuk memastikan kemurnian dari senyawa tetrapeptida tersebut dilakukan proses RP-HPLC yaitu hasil yang didapat pada menit ke 9,243 menunjukkan puncak waktu retensinya. Pada pengujian aktivitas antioksidan dengan menggunakan metode DPPH, hasil yang didapat dari senyawa tetrapeptida (Ser-Pro-Lys-Thr) yaitu nilai inhibisi sebesar 45% yang menunjukan bahwa aktivitas antioksidan dari senyawa tetrapeptida tersebut lemah.Kata kunci : Antioksidan, tetrapeptida, sintesis peptida fase padat
