dc.description Abstract. Gly-Phe-Gly-Cys (GFGC) linear tetrapeptides have been successfully synthesized by the Solid Phase Peptide Synthesis (SPPS) method using a solid phase buffer 2-chlorotrityl chloride resin. The use of Fmoc protected amino acids was chosen in this study. In the preparation of linear tetrapeptide ingredients, a combination of HBTU and HOBt coupling reagent is used. Chlorine test was carried out to ensure the success of amino acid coding and Fmoc  deprotection. After obtaining the arrangement of linear tetrapeptide fragment in the resin, a triflouroacetat (TFA) solution was added to the aquadest to release the resin from the GFGC linear tetrapeptide. The linear tetrapeptide obtained was evaporated using a rotary evaporator and analyzed using a Mass Spectrometer instrument marked by the emergence of molecular ion peaks [M-H]+ m/z 647,236 for GFGC linear tetrapeptide compounds which have side protective groups and molecular ion peaks [M-H]+ m/z 383,151 for GFGC linear tetrapeptide without side protective groups and their purity was seen using a Reverse Phase High Performance Liquid Chromatography (RP-HPLC) instrument characterized by the appearance were still impure and needed to be purified.Keywords: Peptide synthesis, GFGC tetrapeptide, Solid Phase Peptide Synthesis (SPPS), AntibacterialAbstrak. Tetrapeptida linier Gly-Phe-Gly-Cys (GFGC) telah berhasil disintesis dengan metode Solid Phase Peptide Synthesis (SPPS) menggunakan suatu penyangga fase padat yaitu resin 2-klorotritil klorida. Penggunaan asam amino terlindung Fmoc dipilih pada penelitian ini. Pada penyusunan fargmen tetrapeptida linier digunakan suatu reagen pengkopling kombinasi HBTU dan HOBt. Dilakukan uji kloranil untuk memastikan keberhasilan pengkoplingan asam amino dan deproteksi Fmoc. Setelah diperoleh susunan fragmen tetrapeptida linier pada resin, ditambahkan larutan triflouroasetat (TFA) dalam aquadest untuk pelepasan resin dari tetrapeptida linier GFGC. Tetrapeptida linier yang diperoleh dievaporasi menggunakan rotary evaporator dan dianalisis menggunakan instrumen Spektrometer Massa yang ditandai dengan munculnya puncak ion molekul [M-H]+ m/z 647,236 untuk senyawa tetrapeptida linier GFGC yang memiliki gugus pelindung samping dan puncak ion molekul [M-H]+ m/z 383,151 untuk tetrapeptida linier GFGC tanpa gugus pelindung samping serta dilihat kemurniannya menggunakan instrumen Reverse Phase High Performance Liquid Chromatography (RP-HPLC) yang ditandai dengan munculnya kromatogram dengan banyaknya puncak-puncak minor yang menandakan tetrapeptida linier GFGC masih belum murni dan perlu dilakukan pemurnianKata Kunci: Sintesis peptida, tetrapeptida GFGC, Solid Phase Peptida Synthesis (SPPS), Antibakteri
