Abstract. Diarrhea is a condition characterized by the consistency of soft and liquid stools and an increase in the frequency of defecation more than 3 times a day accompanied by or without mucus even bleeding stools. Robusta coffee husk is known to have secondary metabolite compounds that have the potential to be antidiarrheal. In vitro robusta coffee skin research shows that robusta coffee skin extract can kill the bacteria that cause diarrhea. This study aims to determine the antidiarrheal effect of Robusta coffee skin extract on male Swiss Webster mice. Extraction by reflux method using ethanol 96%. This research was conducted with 2 methods, namely protection of diarrhea against Castor Oil with parameters: frequency of defecation, stool consistency, and stool weight and intestinal transit method by calculating the ratio of intestinal distance traveled by a marker to the overall length of the intestine in mice. In this study using 3 doses of 30; 60; and 120 mg/kg, the results are processed using statistical tests. the results of the research, it has been proven that the ethanol extract of Robusta coffee peel gives an antidiarrheal effect as seen from the reduction of intestinal peristalsis in the intestinal transit method and reduced fluid secretion from the intestine causing decreased defecation frequency, fecal consistency becomes denser and the resulting stool weight decreases in the protection method Castor Oil and not significantly different from the comparison group.Keywords: Robusta coffee husk, antidiarrheal, Coffea canephora Pierre ex A.Froehner. Abstrak. Diare merupakan keadaan yang ditandai dengan konsistensi tinja yang lembek bahkan cair dan bertambahnya frekuensi defekasi lebih dari 3 kali dalam sehari disertai dengan atau tanpa lendir bahkan berdarah. Kulit buah kopi robusta diketahui memiliki senyawa metabolit sekunder yang berpotensi sebagai antidiare. Pada penelitian kulit kopi robusta secara invitro menunjukkan bahwa ekstrak kulit kopi robusta dapat membunuh bakteri penyebab diare. Penelitian ini bertujuan untuk mengetahui efek antidiare ekstrak kulit buah kopi robusta pada mencit Swiss Webster jantan. Ekstraksi dengan metode refluks menggunakan pelarut etanol 96%. Penelitian ini dilakukan dengan 2 metode yaitu metode proteksi diare terhadap Oleum ricini dengan parameter frekuensi defekasi, konsistensi feses, dan bobot feses dan metode transit intestinal dengan menghitung rasio jarak usus yang ditempuh oleh marker terhadap panjang usus keseluruhan pada mencit. Pada penelitian ini menggunakan 3 dosis yaitu 30;60; dan 120 mg/kgBB, hasilnya diolah menggunakan uji statitistik. Dari hasil penelitian telah terbukti bahwa ekstrak etanol kulit buah kopi robusta memberikan efek antidiare dilihat dari berkurangnya gerakan peristaltik usus pada metode transit intestinal dan berkurangnya sekresi cairan dari usus menyebabkan frekuensi defekasi menurun, konsistensi tinja menjadi lebih padat dan bobot feses yang dihasilkan menurun pada metode proteksi oleum ricini dan tidak berbeda bermakna dengan kelompok pembanding.Kata Kunci: Kulit buah kopi robusta, antidiare, Coffea canephora Pierre ex A.Froehner. |
