Abstract. Diarrhea is the most common digestive disorder for cats which can cause cats to become dehydrated until death. Escherichia coli is one of the infectious agents that be able to cause diarrhea to cats. Earthworms are known to have a bioactive component, which is lumbricin the one of bioactive components that has antibacterial activity. This research aimed to determine the antibacterial activity potential of earthworm flour and make cat food preparations containing earthworm flour. Earthworm flour was tested by in vitro, antibacterial activity test used the agar diffusion with cup plate technique. The results of the antibacterial activity test showed the formation of inhibitory zones at concentrations of 5, 10 and 15% against Escherichia coli bacteria, with a minimum inhibitory concentration (MIC) at 5% concentration. The best formula for a cat food base contained 40% chicken meat, 30% tuna and 10% tempeh. The best cat food formula is the formula that has been added by 5% concentration of earthworm flour. The formulated cat food is cornet form or semi-solid, brown in color, has a distinctive odor to earthworm flour and has an optimum storage temperature at 4°C. Cat food preparation were stated to have good physical stability based on organoleptic test and storage stability test.Keywords: Earthworm flour (Lumbricus rubellus Hoffmeister), Antibacterial, MIC, Cat food.Abstrak. Diare merupakan gangguan pencernaan paling umum pada kucing yang dapat menyebabkan kucing mengalami dehidrasi hingga sampai kematian. Bakteri Escherichia coli salah satu agen infeksi yang dapat menyebabkan diare pada kucing. Cacing tanah diketahui memiliki komponen bioaktif, yaitu lumbricin yang mempunyai aktivitas antimikroba. Penelitian ini bertujuan untuk menentukan potensi aktivitas antibakteri dari tepung cacing tanah dan membuat sediaan pakan kucing mengandung tepung cacing tanah. Tepung cacing tanah dilakukan pengujian aktivitas antibakteri secara in vitro menggunakan metode difusi agar teknik sumuran. Hasil pengujian aktivitas antibakteri menunjukkan terbentuknya zona hambat pada konsentrasi 5, 10 dan 15% terhadap bakteri Escherichia coli, dengan konsentrasi hambat minimum (KHM) pada konsentrasi 5%. Formula terbaik basis pakan kucing mengandung daging ayam 40%, ikan tongkol 30% dan tempe 10%. Formula terbaik sediaan pakan kucing dengan penambahan tepung cacing tanah pada konsentrasi 5%. Sediaan pakan kucing yang dihasilkan berbentuk kornet atau setengah padat, berwarna coklat, berbau khas tepung cacing tanah dan memiliki suhu optimum penyimpanan pada suhu 4°C. Sediaan dinyatakan memiliki stabilitas fisik yang baik berdasarkan pengujian organoleptis dan stabilitas penyimpanan.Kata Kunci: Tepung cacing tanah (Lumbricus rubellus Hoffmeister), Antibakteri, KHM, Pakan kucing
