ABSTRACT: Antioxidant peptides are a group of peptides that can be used as free radical scavengers, can prevent degenerative diseases and prevent premature aging, one of a combination of natural antioxidant peptides that have been discovered previously is a discovery related to Gly-Ala-Trp-Ala tetrapeptide (GAWA)  which can be used from sardinella aurita fish species of ray-finned fish in the genus sardinella that was found in the Atlantic Ocean and has antioxidant reserves. The synthesis of Gly-Ala-Tra-Leu (GAWL) tetrapeptide was carried out by using the solid phase peptide synthesis method (SPPS) in 2-chlorotrityl chloride resin. The strategy of using Fmoc as an amino acid protective group was chosen in this study. In the preparation of peptide fragments the combination of HBTU and HOBt is used as a coupling reagent in GAWL synthesis. After the peptide fragments are arranged in the resin, a triflouroacetic acid (TFA) combination is added to the dichloromethane (DCM) to release the peptide from the resin then concentrations check using a rotary evaporator.  The purity was analyzed using the Reverse Phase High Performance Liquid Chromatography (RP-HPLC) instrument and appeared at a retention time of 17.409 minutes and it was characterized using a mass spectrometer marked with [M-H] + ion molecules at m / z 44.23 for linear GAWL tetrapeptide compoundsKeywords: Tetrapeptida GAWL, Solid Phase Peptide Synthesis (SPPS) ABSTRAK: Peptide antioksidan merupakan kelompok peptide yang berperan karena dapat penangkap radikal bebas, sehingga dapat mencegah penyakit degeneratif dan menjegah penuaan dini, salah satu senyawa peptide antioksidan alami yang telah ditemukan peneliti sebelum nya yaitu senyawa tetrapeptide Gly-Ala-Trp-Ala (GAWA) yang diisolasi dari ikan sardinella aurita spesies ikan bersirip sinar dalam genus sardinella yang di temukan di Samudra atlantik dan dilaporkan memiliki aktifitas antioksidan yang baik. Telah dilakukan sintesis Tetrapeptida Gly-Ala-Tra-Leu (GAWL) dengan menggunakan metode Solid phase peptide synthesis (SPPS) pada resin 2-klorotritil klorida. Strategi pemakaian Fmoc sebagai gugus pelindung asam amino dipilih pada penelitian ini. Pada penyusunan fragmen peptida digunakan kombinasi HBTU dan HOBt sebagai reagen pengkopling dalam sintesis GAWL. Setelah fragmen peptida tersusun pada resin, ditambahkan kombinasi asam triflouroasetat (TFA) dalam diklorometana (DCM) untuk proses pelepasan peptida dari resin. Dilakukan pemekatan menggunakan rotary evaporator. Dan  dianalisis kemurniannya menggunakan instrumen Reverse Phase High Performance Liquid Chromatography (RP-HPLC) dan muncul pada waktu retensi 17,409 menit. Serta dikarakterisasi menggunakan spektrometer massa yang ditandai dengan dengan munculnya ion molekul [M-H]+ pada m/z 44,23 untuk senyawa tetrapeptide GAWL linier Kata Kunci: Tetrapeptida GAWL, Solid Phase Peptide Synthesis (SPPS)
