Abstract. A burn is damage to the skin layer caused by a hot object. Aloe vera gel and chitosan gel are known to be potentially used as a burn wound healing. This study aims to develop aloe vera gel formula on the basis of chitosan gel which has characteristics and good physical stability and to know the burn wound healing activity of the gel preparations at various concentrations of aloe vera gel. The research was begun with chitosan gel base optimization at concentrations of 2, 3, and 4%. Furthermore, gel preparations were made at optimum chitosan gel concentration and aloe vera gel added at various concentrations of 10%, 20%, and 30%. The gel preparations were subjected to a physical evaluation and activity test for the healing of burns on Swiss Webster male mice. The test was carried out for 21 days and observed the test parameters including the time of formation and removal of the scab and the diameter of the burn. The results showed that gel preparations with 4% chitosan concentration in the three concentrations of aloe vera gel had good physical properties based on organoleptic testing, homogeneity, viscosity, flow properties, pH, dispersibility and stability. Gel preparations with aloe vera gel concentrations of 20% and 30% had significant burn healing activity with control preparations (p <0.05). Gel preparations with aloe vera gel concentration of 30% have a better burn healing activity compared to the comparative preparations (p <0.05).Keyword: Gel, burn, aloe vera, chitosan, mice. Abstrak. Luka bakar adalah suatu kerusakan lapisan kulit yang disebabkan oleh benda panas. Gel lidah buaya dan gel kitosan diketahui berpotensi digunakan sebagai penyembuh luka bakar. Penelitian ini bertujuan untuk mengembangkan formula gel lidah buaya dalam basis gel kitosan yang memiliki karakteristik dan stabilitas fisik yang baik serta mengetahui aktivitas penyembuh luka bakar dari sediaan gel tersebut pada berbagai konsentrasi gel lidah buaya. Penelitian diawali dengan optimasi basis gel kitosan pada konsentrasi 2, 3, dan 4%. Selanjutnya sediaan gel dibuat pada konsentrasi gel kitosan optimum dan ditambahkan gel lidah buaya pada berbagai konsentrasi 10%, 20% dan 30%. Terhadap sediaan gel dilakukan evaluasi fisik dan uji aktivitas penyembuh luka bakar terhadap mencit jantan galur Swiss Webster. Pengujian dilakukan selama 21 hari dan diamati parameter uji meliputi waktu terbentuk dan terlepasnya keropeng serta diameter luka bakar. Hasil penelitian menunjukkan bahwa sediaan gel dengan konsentrasi kitosan 4% pada ketiga konsentrasi gel lidah buaya memiliki sifat fisik yang baik berdasarkan pengujian organoleptis, homogenitas, viskositas, sifat alir, pH, daya sebar dan stabilitas. Sediaan gel dengan konsentrasi gel lidah buaya 20% dan 30% memiliki aktivitas penyembuh luka bakar yang berbeda bermakna dengan sediaan kontrol (p<0,05). Sediaan gel dengan konsentrasi gel lidah buaya 30% memiliki aktivitas penyembuh luka bakar yang lebih baik dibandingkan dengan sediaan pembanding (p<0,05).Kata kunci : Gel, luka bakar, lidah buaya, kitosan, mencit. |
